Miranda's loyalty mission is The Prodigal. Bear in mind, though, that doing the mission won't gaurantee a romance.
Once you've obtained the initial teammember dossiers and recruited all the members, you'll reach a forced mission. Afterward your teammembers will begin to want to get some personal issues out of the way. Kelly, your yeoman, will let you know if any team member wants to talk to you. Miranda and jacob are the first members with missions. After talking with Miranda, and agreeing to help her out, you need to go to Ilium and talk to her. She's in the bar. Just complete the mission and you'll have her loyalty, for now.
From here on out you need to at least side with Miranda. If a dispute occurs you should side with her unless you can reach a diplomatic solution. Just keep talking to her, complimenting her and eventually you can flirt a little. She'll flirt back. Once she says that she'll come to you when things get quieter, you're set. Once you head through the Omega-4 relay the magic happens.

Miranda: The Prodigal
She tells you about her sister when YOU go to speak to her. Even then you have to choose the correct dialouge and be a male shepard for the scene. Before or after the final story mission I think you have to go to your rooms terminal and call her up.I haven't gotten any relationships completed yet but the reason I say "after" is because the game, depending on if you survive, let's you finish the other missions instead of ending the game right there.
